In the polymerase chain reaction, which step requires the highest temperature?
Answer and explanation
Correct answer: A. Denaturation of the template DNA
- Step 1: Denaturation at about 95 degrees Celsius separates the DNA strands. Step 2. Annealing is at about 55 and extension at about 72 degrees. Answer: denaturation. Why not the others: annealing and extension occur at lower temperatures, and PCR has no ligation step.
